    以老芒麦(Elymus sibiricus)种子为材料,采用高温(45℃)、高湿(100%RH)的方法处理供试种子,测定电导率、丙二醛、可溶性糖、可溶性蛋白的含量,SOD和POD活性等相关生理生化指标,并采用酸性聚丙烯酰氨凝胶电泳(A-PAGE)技术对不同程度老化种子的醇溶蛋白组成进行分析,研究人工老化对老芒麦种子生理生化特性及醇溶蛋白组成的影响。结果表明:随着人工老化程度的加重,种子浸出液电导率和MDA 含量都呈现逐渐增加的趋势;可溶性蛋白含量呈现先上升后下降的趋势,老化3 d后,可溶性蛋白含量急剧下降,老化7 d后,可溶性蛋白含量与对照相比下降了11.89 mg/g;老化处理7d后,可溶性糖含量与对照相比增加,增幅为79.05%;随着老化程度的加剧,SOD、POD 活性都逐渐下降,老化7 d后,SOD和POD活性依次下降44.49%、32.62%;SOD活性下降大于POD活性下降。种子在老化过程中醇溶蛋白发生了一系列的变化,醇溶蛋白图谱颜色在处理的第3 d开始变浅,且随着老化时间的延长,α,β 和ω 区醇溶蛋白出现丢失的现象。

    Abstract:

    Elymus sibiricus seeds treated with high temperature (45℃) and high humidity (100% RH)were used as materials.The changes of physiological and biochemical indexes (electrical conductivity,malondialdehydecontent,soluble sugar content,soluble protein content,SOD activity and POD activity) were measuredand the constitute of gliadin was analyzed by acid-soluble polyacrylamide gel electrophoresis (A-PAGE) technique.The results showed that the conductivity and MDA content of seed showed a gradual increase trend withthe increase of artificial aging degree;The soluble protein content showed a trend of increasing first and then decreasingand decreased sharply after aging for 3 days.Compared with the control,the content of soluble proteindecreased by 11.89 mg/g after aging for 7 days.Compared with the control,the content of soluble sugar increasedby 79.05% after aging treatment for 7 days;The activity of SOD and POD decreased gradually with theincrease of artificial aging degree.The activity of SOD and POD decreased by 44.49% and 32.62% after agingfor 7 days,respectively.The degree of SOD activity decreased was greater than that of POD activity (44.49%>32.62%) after aging treatment for the same time.During the aging process,the constitute of gliadin waschanged.The color of gliadin band lightened on the third day and some gliadin bands disappeared in α,β,ω regionwith the increase of artificial aging degree
