以4个紫花苜蓿品种Maverick(FD1对照,秋眠型苜蓿,秋眠1级)、Vernal(FD2,秋眠型苜蓿,秋眠2级)、UC-1887(FD10,极不秋眠型苜蓿,秋眠10级)和UC-1465(FD11,极不秋眠型苜蓿,秋眠11级)为材料,采用SYBR-Green I qPCR来计算基因表达定量,检测其在不同生长状态下,光受体PHYA、PHYB、CRY2B、FHY1基因表达量,揭示不同光受体的基因表达量变化规律。结果表明:光敏色素PHYA和PHYB在调控苜蓿秋眠与生长时具有协同作用;蓝光受体CRY2B对秋眠的反应灵敏度强于其热应激的反应,且PHYA,PHYB和CRY2B都参与了对苜蓿秋眠的调控,它们之间存在互作效应。
Abstract:
Fall dormancy is a growth characteristic of alfalfa.It is a special dormancy way of alfalfa to copewith environmental changes,which related to the length of sunshine and the sensitivity of photoperiod.We detectedthe gene expression of PHYA,PHYB,CRY2B and FHY1 of Alfalfa with different fall dormancy varietiesin different growth states using QPCR SYBR-Green I in this experiment,which the varieties were fall-dormantvariety Maverick(FD1),Vernal(FD2),extremely non-dormant variety UC-1887(FD10) ,and UC-1465(FD11).The regulation of light receptor gene expression could provide theoretical support for the regulation mechanismof Alfalfa fall dormancy.The results showed that the photosensitive PHYA and PHYB had synergistic effect onthe regulation of Alfalfa fall dormancy and growth.The sensitivity of blue light receptor CRY2B to fall dormancywas stronger than that of its heat stress.PHYA,PHYB,and CRY2B participated in the regulation of Alfalfafall dormancy,and there was an interaction effect between them.
