    以紫花苜蓿品种瑞诺(Medicago sativa cv.Creno)为供试种样,采用涂布法和平板划线法对紫花苜蓿种带耐盐细菌进行纯化分离,在不同浓度梯度的含盐培养基中进行培养,分离得到4株种带耐盐细菌,分别为RN1、RN2、RN3、RN4,其耐盐性分别为3% NaCl、5% NaCl、5% NaCl、9% NaCl。结合表型特征和16S rDNA 基因序列,初步鉴定为芽孢杆菌属(Bacillus sp.)、欧文菌属(Erwinia sp.)、芽孢杆菌属(Bacillus sp.)和葡萄球菌属(Staphylococcus sp.)

    Abstract:

    By exploring the diversity of salt-tolerant bacteria in lucerne seed,a number of salt-tolerant bacteriaresources were screened out in this study.Medicago sativa var.creno was selected as the test sample.Salttolerantseed-borne bacteria of alfalfa were isolated and purified by dilution coating method and plate markingmethod.Four salt-tolerant bacteria(RN1,RN2,RN3 and RN4) were isolated in saline medium with differentconcentration gradients and their salt-tolerance was 3% NaCl,5% NaCl,5% NaCl and 9% NaCl respectively.Combined with the phenotypic characteristics and 16S rDNA gene sequence,they were identified as genus membersof Bacillus sp.,Erwinia sp.,B.sp.and Staphylococcus sp.This study provides bacterial resources for furtherdevelopment and utilization of functional bacteria suitable for saline-alkali land improvement.
