    【目的】利用生物信息学方法,鉴定草地早熟禾(Poa pratensis)胚胎发生标记基因 PpBBM, 并通过保守基序比对和进化树分析,推测可能具有孤雌生殖功能的 PpBBM 基因。【方法】基于草地早熟禾转录组数据库信息,检索 PpBBM 基因。通过生物信息学软件分析 PpBBMs 蛋白理化特性,比较不同 PpBBMs 蛋白的系统进化关系和保守基序差异 。【结果】共获得 3 个具有完整开放阅读框的 PpBBMs 基因,其编码的蛋白均含有 2 个 AP2 结构域,其中,PpBBM1 和 PpBBM2 具有相同的 motif 类型和数量。3 个 PpBBMs 蛋白都是酸性蛋白,定位于细胞核,不存在跨膜结构和信号肽。系统进化分析表明,30 个物种的 BBM/BBML 蛋白成员共聚为 5 类。其中,PpBBM1、PpBBM2 聚在第 2 分支,分别与 BdBBM 和 SbBBML 亲缘关系最近 ;PpBBML 位于第 5 分支 ,与 ZmBBML 亲缘关系最近 。 PpBBM1、PpBBM2 和 PpBBML 均具有 BBM 基因所特有的 bbm-1 基序以及 AP2 亚家族特定的 euANT2-4 基序,但 PpBBML 蛋白的所有基序均有个别氨基酸残基存在变异。【结论】与 PpBBML 基因相比,PpBBM1 PpBBM2 更有可能具有诱导孤雌生殖的功能。

    Abstract:

    【Objective】 Using bioinformatics methods,this paper aims to identify the embryogenic marker gene PpBBM of kentucky bluegrass and predict its parthenogenetic function through conservative motif alignment and evolutionary tree analysis.【Method】 Based on the information from the Transcriptome database of kentucky bluegrass,the PpBBM gene was searched. The physical and chemical properties of PpBBMs protein were analyzed,and the phylogenetic relationship and conservative motif differences of PpBBMs proteins were compared through bioinformatics software.【Result】 A total of three PpBBMs genes with complete open reading frame were obtained,and the proteins encoded by them all contained two AP2 domains, among which PpBBM1 and PpBBM2 have the same motif type and number. The three PpBBMs proteins are acidic proteins located in the nucleus without transmembrane structure and signal peptide. Phylogenetic analysis showed that the menbers of BBM/BBML prdens of 30 species were polymerized into five groups. Among them,PpBBM1 and PpBBM2 are clustered in the second branch and have the closest genetic relationships with BdBBM and SbBBML,respectively;PpBBML is located in the fifth branch and has the closest genetic relationship with ZmBBML. PpBBM1,PpBBM2,and PpBBML all have the bbm ‐1 motif unique to the BBM gene and the euANT2‐4 motif specific to the AP2 subfamily. However,all motifs of the PpBBML protein exhibited variations in individual amino acid.【Conclusion】PpBBM1 and PpBBM2 genes are more likely to have the ability to induce parthenogenesis compared with PpBBML.
