    摘要:

    【目的】 探讨马蔺(Iris lactea)不同种质花瓣抗氧化能力的差异,筛选出花瓣抗氧化能力强的马蔺种质资源,为马蔺种质资源开发利用奠定基础。【方法】 选取我国6 个省区不同生境条件下22 份马蔺种质资源,对其盛花期花瓣的花青素、类黄酮、总酚、原花青素、羟自由基清除率及总抗氧化能力进行测定,并进行相关性分析,同时采用主成分分析法和隶属函数法对各种质材料的抗氧化能力进行综合评价。【结果】 1)22 份马蔺种质间各成分含量存在一定差异,其中原花青素含量变异系数最大,为64. 41%,总抗氧化能力含量变异系数最小,仅为4. 53%;花青素含量为0. 15~0. 56 mg/g,类黄酮含量为0. 51~1. 34 mg/g,总酚含量为1. 19~1. 90 mg/g,原花青素含量为0~0. 19 mg/g;羟自由基清除率在24. 97%~50. 82%,总抗氧化能力为1. 98~2. 36 μmol Trolox/g;2)测定的6 项生理生化指标间均存在不同程度的相关性,且根据欧氏距离聚类分析的结果,可将参试的22 份马蔺种质资源抗氧化能力聚为3 大类,包括抗氧化能力强的8 份(ML011、ML012、ML013、ML014、ML016、ML019、ML029 和ML035),抗氧化能力中等的9 份和抗氧化能力弱的5 份; 3)马蔺种质抗氧化能力的综合评价排序结果为ML014 > ML013 > ML011 > ML012 > ML035 >ML19>ML016>ML029>ML018>ML008>ML010 > ML006 > ML020 > ML005 > ML009 > ML007 > ML023>ML017>ML032>ML015>ML004>ML001,与聚类分析结果高度一致。【结论】 22 份马蔺种质材料中ML011、ML012、ML013、ML014、ML016、ML019、ML029 和ML035 抗氧化能力强,为优异马蔺种质资源的挖掘及产品开发利用提供科学依据。

    Abstract:

    【Objective】 In order to explore the differences in antioxidant capacity of petals of various germplasmmaterials of Iris lactea Pall. var. chinensis( Fisch.) Koidz. ,the accessions of I. lactea with strong antioxidant capacitywere screened out,which would lay a foundation for the evaluation,development and utilization of Iris lactea accessions.Method In this paper,22 I. lactea accessions under different habitat conditions in 6 provinces of China wereselected to determine anthocyanin,flavonoid,total phenol,proanthocyanidin,hydroxyl radical scavenging rate and totalantioxidant capacity of petals at full flowering stage,and the correlation between each index was analyzed. At thesame time,the antioxidant capacity of each material was comprehensively evaluated by principal component analysiscombined with membership function method.【Result】 (1) The results showed that there were some differences inthe content of each component among 22 I. lactea accessions. Among them,the coefficient of variation of proanthocyanidincontent was the largest (64. 41%),and the coefficient of variation of total antioxidant capacity content was thesmallest (4. 53%). The anthocyanin content of the 22 samples was 0. 15~0. 56 mg/g,the flavonoid content was0. 51~1. 34 mg/g,the total phenol content was 1. 19~1. 90 mg/g,the proanthocyanidin content was 0~0. 19 mg/g,and the hydroxyl radical scavenging rate ranged from 24. 97 % to 50. 82 % . The total antioxidant capacity was between1. 98 μmol Trolox/g and 2. 36 μmol Trolox/g.(2) The results of correlation analysis showed that there weredifferent degrees of correlation among the six physiological and biochemical indexes. According to the results of Euclideandistance cluster analysis,the antioxidant capacity of 22 I. lactea accessions could be clustered into three categories.,of which 8 had strong antioxidant capacity,including ML011,ML012,ML013,ML014,ML016,ML019,ML029 and ML035,9 had moderate antioxidant capacity and 5 had weak antioxidant capacity.(3) The results ofcomprehensive comparison of antioxidant capacity of I. lactea accessions by principal component analysis combinedwith membership function method were ML014 > ML013 > ML011 > ML012 > ML035 > ML19 > ML016 >ML029 > ML018 > ML008 > ML010 > ML006 > ML020 > ML005 > ML009 > ML007 > ML023 >ML017 > ML032 > ML015 > ML004 > ML001,which were highly consistent with the results of cluster analysis.【Conclusion】 The above research shows that ML011,ML012,ML013,ML014,ML016,ML019,ML029 andML035 have strong antioxidant capacity in 22 I. lcateaaccessions,which provides scientific basis for the excavation ofexcellent Iris germplasm resources and the development and utilization of products.
